Chitosan-Based Particulates Carriers for Pulmonary Drug Delivery

Chitosan (CH), a naturally occurring linear polysaccharide, is a well-known carrier for drug delivery due to its biodegradable and biocompatible nature. Additionally, CH and its synthetic derivatives have been widely explored as cargos in nano- and microparticulate drug delivery systems for different routes of administration owing to its distinct, pH sensitivity, low toxicity, and controlled drug release properties. It has also been established as a pulmonary drug carrier in the therapy of various infectious diseases. The success of CH and its synthetic derivatives as a pulmonary drug carrier is a vital interplay of their mucoadhesive nature, permeation properties, and site-specific delivery potential. In the present chapter, we have systematically reviewed the pulmonary applications of CH-based novel drug delivery systems considering their aerosolization potential and in vivo respiratory dispersion performance. Moreover, different studies discussing CH cytotoxicity potential with the help of in vitro human cell lines are also elaborated. Lastly, a few critical concerns linked to the applicability of CH carriers in the real world are proposed to provide insights for future investigations.
